利用大肠杆菌菌株表达出高纯度的酸性成纤维细胞生长因子,并对其促3T3细胞的增殖作用和对实验秃毛大鼠的疗效进行了研究。结果表明,在体外实验中,1.95ng/ml~1000ng/ml的aFGF溶液可以促进balb/c3T3细胞的分裂增殖,与PBS组比较具有显著性差异(P〈0.05,P〈0.01);在体内实验中,在第7天,4μg/ml aFGF组大鼠毛发长度变长,与模型对照组比较有显著性差异(P〈0.05);在第14天,2μg/ml和4μg/ml aFGF组大鼠毛发长度继续变长,与模型对照纽比较有显著性差异(P〈0.05)。病理检查结果显示aFGF可以促进实验秃毛大鼠的毛囊数目增多,毛囊无萎缩变小现象,血管无充血现象,基本恢复到正常水平。由此得出aFGF可以促进3T3细胞的分裂增殖,以及促进实验秃毛大鼠的毛发生长。aFGF具有很好的开发前景。
Escherichia coli was used to produce acidic fibroblast growth factor(aFGF) of high purity, and its effect on proliferation of 3T3 and improvement of bald rats were studied. The result shows that in vitro aFGF at the level from 1.95 ng/ml to 1000ng/ml can promote the proliferation of balb/c 333, which has significant difference when compared with the control group(P 〈0.05 ,P 〈0.01 ). In vivo aFGF at the level of 2μg/ml and 4μg/ml can improve the bald hair of the rats, which also has significant difference when compared control group (P 〈 0. 05). Pathology result proves that aFGF can improve the hair numbers of the bald well as the hyperaemia situation of the veins. It indicates that aFGF has good future in clinical research.
