目的研究非促分裂人酸性成纤维细胞生长因子(nm-haFGF)对H2O2损伤的人视网膜色素上皮(RPE)细胞的保护作用。方法利用H2O2建立人RPE细胞氧化损伤模型,采用MTT法检测RPE细胞的存活率,核染色观察RPE细胞的核形态,流式细胞仪检测RPE细胞死亡率以及Western blot检测bcl-2的表达。结果nm-haFGF预处理可以提高H2O2损伤的REP细胞的存活率;nm-haFGF质量浓度在0-10mg/L的范围内,RPE细胞存活率从67.4%上升至92%;nm-haFGF刺激后,RPE细胞的bcl-2表达增加。结论nm-haFGF对H2O2氧化损伤RPE的细胞具有保护作用,并在一定范围内呈质量浓度依赖性。其保护机制可能是通过bcl-2的增加而产生抗凋亡作用。
Objective RPE cell is subject to injury by reactive oxygen species (ROS).Studies have demonstrated that nm-haFGF can prevent organs and tissues damage from oxidative stress.The purpose of this study was to study the protective effects and its possible mechanism of non-mitogenic human fibroblast growth factor(nm-haFGF)on human RPE cell injury induced by hydrogen peroxide.MethodsHuman retinal pigment epithelium (RPE)cell strains were cultured in RPMI1640 containing 10% bovine serum.The oxidative injury model of RPE cell was induced by the utilization of 0,0.1,0.2,0.4,0.8,1.0mmol/L H2O2,respectively.Part of RPE cells was pretreated using 0,0.01,0.05,0.10,0.25,0.50,1.00,2.50,5.00,10.00mg/L of nm-haFGF and then 0.4mmol/L H2O2 was added in medium.The survival rate of the cells in different concentrations of H2O2 was detected using MTT to evaluate the cytotoxic effects of H2O2.The apoptotic cells were identified by Hoechst 33342 staining,and death of the cells was detected by flow cytometry (FCM).Expression of bcl-2 in cultured RPE cells was detected by Western blot analysis.ResultsThe survival rate of the cells was significant different among different concentrations of H2O2 group(F=182.794,P=0.000).Viability rate of the cells was gradually reduced with the increase of H2O2 concentration,showing statistically significant differences in comparison with control group(P〈0.05-0.01).The RPE cells pretreated by nm-haFGF showed a higher survival rate as the increase of concentration of nm-haFGF(F=13.072,P=0.000)and presented considerable differences in various concentrations of nm-haFGF compared with H2O2 group (P〈0.05-0.01).The viability of the cells increased from 67.4% to 92% as the increase of nm-haFGF concentration.Nm-haFGF pretreatment up-regulate bcl-2 of RPE injury induced by H2O2.ConclusionNm-haFGF can prevent RPE cells from injury induced by H2O2 in concentration-dependent manner.
