目的:建立一种操作简单、可行的大鼠异位心肺联合移植模型,以利于移植排斥反应机制的研究及其实验性治疗研究。方法:分离受体Wistar大鼠颈部并形成皮囊,提供容受供体心肺的空间,分离颈总动脉。供体经过肝素化后,在0~4℃条件下取心肺,并作适当修剪,最后在0~4℃条件下将心肺供体SD大鼠的主动脉与受体Wistar大鼠的颈总动脉行端侧吻合,并进行心肺复苏。结果:心肺联合移植正式手术50例,手术后通血复跳率96%。手术时间为(70±10)min,平均冷缺血时间为(40±10)min。手术主要并发症是动脉吻合口狭窄及出血,血栓形成,失血性休克。未作任何处理的移植心肺3天存活率82%。结论:成功建立了大鼠颈部心、肺联合移植模型。
Objective: To establish a simplified and useful heterotopy heart-lung transplantation rat model for studying of the mechanism of transplantation rejection and cure the rejection in animal model. Methods: The heterotopic cervical heart-lung transplantation model was established by anastomosing the aorta of the SD rat heart to the right common carotid of the Wistar rat. A total of 50 couples allogene rats were performed same proceeding like that. Results : The rate of successful beat of the donors' heart was 96%. The average operative time and graft ischemia time were ( 70 ± 10) min and (40 ± 10) min respectively. The main complications included hemorrhage and stenosis of anastomosing stoma, thrombosis and hemorrhagic shock. The 3 days survival rate of heart-lung allograft without any treatment was 82%. Conclusion: A simplified and useful rat heterotopic heart-lung transplantation model was established successfully.
