目的:探讨β-1,4-半乳糖基转移酶Ⅰ与雪旺细胞衰老的相关性。方法:以不同浓度的β-1,4-GalT—Ⅰ表达质粒转染雪旺细胞,观察衰老相关半乳糖苷酶染色情况。以Ha-Ras转染雪旺细胞诱导衰老,采用Northern blot方法检测转染后不同时间β-1,4-GaIT—Ⅰ表达变化。结果:随着转染β-1,4-GaIT—Ⅰ表达质粒浓度的提高,细胞衰老相关半乳糖苷酶染色阳性率增高;随着Ha—Ras转染时间的延长,β-1,4-GalT—Ⅰ表达逐渐增高。结论:β-1,4-GalT-Ⅰ与雪旺细胞的衰老相关,并可能在周围神经雪旺细胞的增殖调节中发挥重要作用。
Objective: To study the relations between β-1,4-GalT-Ⅰ and the aging of Schwann cells. Methods: The stain of sEnescence-association β-galactosidase was studied in Schwann cells transfected with β-1,4-GalT-Ⅰ expression plasmids of various concentrations. Northern blot was used to investigate the changes of β-1,4-GalT-Ⅰ in Schwann cells transfected with the Ha-Ras expression plasmids of various time.Results: The stain of senescence-association β-galactosidase increased when the concentration of β- 1,4-GalT-Ⅰ expression plasmids increased, and the expression of β-1,4-GalT-Ⅰ mRNA increased with the increased days of transfected Ha-Ras in Schwann cells.Conclusion: It is suggested that some relations exist between β-1,4-GalT-Ⅰ and the aging of Schwann cells, and that may play an important role in regulating the proliferation of Schwann cells in peripheral nerves.
