经多菌株的虫生真菌生物活性筛选,发现一株被毛孢菌株RCEF0851发酵物具有较强的免疫增强作用,进一步采用荧光定量PCR方法研究RCEF0851不同提取物对体外培养小鼠巨噬细胞RAW 264.7的IL-1β、IL-6和TNF-α转录水平的影响。结果表明,乙酸乙酯提取物能显著提高TNF-a转录水平的表达,但对IL-1β和IL-6的转录调节作用不明显;水提物和甲醇提取物能明显提高小鼠巨噬细胞中IL-1β、IL-6和TNF-α的转录水平,表明它们具有免疫增强作用。
In the process of a screening for bioactive metabolites of entomogenous fungi, the fermentation broth of a strain of HirsuteUa sp. RCEF0851 exhibited strong immuno-enhancive properties. Therefore, we launched a systematic studies on immunomodulatory activities of different extracts from mycelia of RCEF0851 by using real time qPCR method. The results showed that the ethyl acetate extract could enhance gene transcription of TNF-α, but had no effect on gene transcription of IL-1β and IL-6, water and methanol extracts could up-regulate gene transcription of IL-1β, IL-6 and TNF-α. This means that the extracts of RCEF0851 have immuno-enhancive activities.
