目的:预测并分析比较衣原体MOMP蛋白的二级结构和B细胞表位。方法:以各株衣原体MOMP蛋白的氨基酸序列为基础,采用Gamier—Robson法、Chou—Fasman法和Karplus-Schulz法预测蛋白二级结构;按Kyte-Doolittle法、Emini法和Jameson—Wolf法预测蛋白的抗原表位;以ClustalX软件序列比对分析其多变区。结果:各株MOMP蛋白含多个抗原位点,各序列在第80—107、165—178、249—264、332—346位序列高度差异,并出现比对“空沟”。多个强表位位于序列保守区内。结论:MOMP蛋白有较强的免疫原性,预测的抗原位点为之后蛋白相互作用研究、单抗制备、亚单位疫苗设计提供了理论依据。
Objective: To predict the secondary structure and B cell epitope of MOMP protein of Chlamydia. Methods: The secondary structure was predicted by the method of Gamier-Robson,Chou-Fasman and Karplus-Schulz, and its cell epitope was predicted by the method of Kyte-Doolittle, Emini and Jameson-Wolf. The diversity region was analysed by ClustalX. Results: There were some cell epitopes in MOMP proteins and the different sections in 80-107, 165-178, 249-264, 332-346. Conclusion: MOMP protein has strong immumogenicity.The research provides the basis for the preparation of monoclonal antibody, the reference for the discussion for the moleular regulation mechanism of protein to protein and the design of subunit vaccine.
