沙眼衣原体E型重组主外膜蛋白疫苗和DNA疫苗免疫效应的比较

目的比较E型重组主外膜蛋白(rMOMP)疫苗和MOMPDNA疫苗诱导小鼠产生的特异性免疫效应.方法用纯化的rMOMP和DNA疫苗分别通过股四头肌肌注和阴道黏膜涂布的途径免疫BALB/C雌鼠。通过检测小鼠血清IgG抗体水平和阴道冲洗液sIgA抗体水平、脾淋巴细胞增殖、迟发型超敏反应、细胞因子检测、阴道脱落细胞种植等指标进行两组免疫效应的比较.结果rMOMP肌注组小鼠:血清IgG抗体水平OD450值为0.424±0.075,迟发型超敏反应小鼠足垫增厚0.272±0.054mm;DNA肌注组:血清IgG抗体水平OD450值为0.332±0.033,迟发型超敏反应小鼠右足垫增厚0.258±0.041mm。通过肌注途径诱发的保护性免疫应答rMOMP蛋白疫苗好于DNA疫苗(P〈0.05)。结论rMOMP蛋白疫苗的免疫应答强于DNA疫苗,通过阴道涂布途径不能诱导小鼠产生特异性体液和细胞免疫应答。

英文摘要:

Objective To compare specific immune responses induced in mouse immunized with the vaccine of rMOMP and MOMP DNA. Method Female BALB/c mouse were immunized intranmscularly and intravaginally with rMOMP and DNA. hnmune responses in the mouse was assayed through detection of the antibody levels of Chlamydia-specific in sera and vaginal washes, lymphocyte proliferation, delayed hypersensitivity and cytokines. Vaginal swabs were collected and cultured for Chlamydia trachomatis after genital challenged and so on. Results The IgG level of Chlamydia-specific in mice immunized intramuscularly with rMOMP is 0. 424±0. 075 (OD450 ), and is 0. 332±0. 033 (0D450 ) in mice immunized intramuscularly with MOMP, The thickness of right hind footpad of mice added 0. 258±0. 041ram in rMOMP treatment group and 0. 258±0.041mm in MOMP DNA treatment group. The immune responses in mice immunized with the rMOMP by intramuscular was better than that immunized with the DNA(P 〈0.05). Conclusion The immune responses induced in mice vaccinated with rMOMP are better than that with DNA. The mice couhln't induce humoral and cellular immune responses to Chlamydia-specific by intravaginal treatment.
