Most cells contain a variety of transport vesicles traveling to different destinations. Although many specific transport routes exist, the basic molecular principles appear to be rather similar and conserved in evolution. Adequate evidence showed that membrane fusion needed the participation of Sec1/Munc18 (SM) proteins besides the SNARE protein family. But, contrary to the uniform clarity of SNARE proteins, different data on SM proteins were obtained in different experimental systems, resulting in researchers holding different views for exact effect and action site of SM proteins and its action mode with SNARE proteins. Different SM proteins and SNARE proteins have three kinds of different action modes, In recent researches,it has been determined that Munc18-1 directly promotes fusion and maybe interacts with SNARE proteins in all three modes. This paper reviewed the advancement of this domain.
