为了解云南省杂鳞库蚊复合组蚊虫的分子分类及COI基因序列特征,2013年5月在云南省江城县采集标本,提取蚊虫基因组DNA,用COI基因特异引物PCR扩增和序列测定,然后采用生物信息学软件进行核苷酸同源性和遗传进化分析。共采集蚊虫标本148只,其中致倦库蚊112只(75.67%)、杂鳞库蚊复合组蚊虫12只(8.11%)、骚扰阿蚊17(11.49%)、其他蚊虫7只(4.73%)。序列分析结果显示,12只库蚊COI基因核苷酸同源性在94.7%~100之间,M1、M3、M4、M5、M8、M9、M10、M11、M12共9只蚊虫与三带喙库蚊位于同一进化分支内,核苷酸同源性高达98%~100%,而M2、M6、M7三只蚊虫与三带喙库蚊核苷酸同源性仅在94.8%~95.9%之间,区别于三带喙库蚊和杂鳞库蚊单独形成一个进化簇。云南省江城县地区存在两种杂鳞库蚊复合组的库蚊。
To understand the molecular taxonomy and characteristics of COI genes of Culex vishnui complex in Yunnan province,2013 May,we collected insects from Jiangcheng county of Yunnan province.The genome DNA of mosquitoes were extracted and COI genes were amplified by using specific primers and sequenced,which were analyzed by using bioinformatics software for nucleotides homology and phylogenetic relationships.In total,we collected 148 insects that include 112 Culex quinquefasiatus(75.67%),12 Culex vishnui complex(8.11%),17 Armigeres subalbatus(11.49%)and 7others(4.73%).The results of sequence analysis showed that the homology of nucleotides between twelve CulexCOI genes is varied from94.7%to 100%.Nine mosquitoes(M1,M3,M4,M5,M8,M9,M10,M11 and M12)and Culex tritaeniorhynchus located in the same cluster and the homology of nucleotides is up to between 98% and 100%.The homology of nucleotides between the three mosquitoes(M2,M6 and M7)and Culex tritaeniorhynchus is varied from 94.8% to 95.9% and there three mosquitoes are clustered into a sole volutionary branch which is distinct fromCulex tritaeniorhynchus and Culex vishnui.In this study,these mosquitoes were identified and classified by molecular biological methods and we concluded that there are two Culex vishnui complexes in Jiangcheng county of Yunnan province.
