目的进行不同产地、不同品种大黄药材中苯丁酮及二苯乙烯类成分的含量测定。方法以HPLC法对大黄药材中反-3,5,4′-三羟基苯乙烯基-4′-O-β-D-葡萄糖苷(a)、4′-羟基苯基-2-丁酮(b)、4′-羟基苯基-2-丁酮-4′-O-β-D-(6″-没食子酰基)-葡萄糖苷(c)等3种成分进行含量测定。流动相为甲醇-1%冰醋酸溶液(20∶80);检测波长280 nm;流速1.0 mL/min;柱温35℃。结果正品大黄药材中均能检测到上述3种成分,成分a在掌叶大黄中的含量普遍高于唐古特大黄和药用大黄,伪品土大黄中含量最低,仅为掌叶大黄的4%左右。成分b和c的含量在掌叶和唐古特大黄中较为接近,药用大黄中含量较低。伪品土大黄中不含成分c。结论 3种成分均可作为大黄药材的质量评价指标,丰富了大黄药材的质量评价方法。
Objective To determine the content of butyrophenone and stilbene constituents in different varieties of medicinal material of Dahuang(Radix et Rhizoma Rhei) produced from different areas.Methods The content of trans-3,5,4′-3-hydroxy-vinyl-4′-O-β-D-glucopyranoside(a),4′-hydroxy-phenyl-2-butanone(b) and 4′-hydroxy-phenyl-2-butanone-4′-O-β-D-(6″galloyl)-glucopyranoside(c) in Dahuang were determined by HPLC at 35℃.The mobile phase was methanol-1% acetic acid water solution(20∶80).The detection wavelength was 280 nm and flow velocity was 1.0 mL/min.Results In identified material of Dahuang three constituents were detected.The content of a was higher in Zhangye Dahuang(Rheum palmatum L.) than that in Tanggute Dahuang(Rheum tanguticum Maxim.ex Balf.) and in medicinal Dahuang.It was the lowest in spurious Tu Dahuang(Rumex madaio Makino) and counted for 4% of that in Zhangye Dahuan.The content of b and c were similar in Zhangye Dahuan and Tanggute Dahuang and lower in medicinal Dahuang.There was no c in spurious Tu Dahuang.Conclusion These three constituents can be taken as the indicators of quality evaluation of Dahuang,which enrich the method of quality evaluation of Dahuang material.
