利用超声回流法获取不同溶剂(水、乙酸乙酯、无水乙醇、丙酮、环己烷)的黄皮叶提取物,选用滤纸片扩散法进行抑菌试验、二倍稀释法测定最小抑菌浓度,研究黄皮叶提取物对十种微生物的抑制作用.结果表明:乙酸乙酯、无水乙醇、丙酮提取物对供试的细菌(金黄色葡萄球菌、大肠杆菌、枯草芽孢杆菌、白色葡萄球菌、四联微球菌、蜡状芽孢杆菌)有明显抑菌效果.乙酸乙酯提取物对大肠杆菌、白色葡萄球菌的最小抑菌浓度为3.2 mg/mL;对金黄色葡萄球菌、蜡状芽孢杆菌、枯草芽孢杆菌的最小抑菌浓度为6.3 mg/mL;对四联微球菌的最小抑菌浓度是12.5 mg/mL.乙酸乙酯提取物抑菌性受温度、pH影响较大,受光照影响不明显.各种溶剂的超声提取物对供试真菌(黑曲霉、青霉、根霉、白色念珠菌)均无抑菌效果.该研究结果为开发利用黄皮叶的防腐、抑菌作用提供参考.
The effective components from Clausena lansium leaves were extracted using the ultrasound reflux. The antimi crobial effects of different solvents (water, ethyl acetate, anhydrous ethanol, acetone, cyclohexane) extracts were observed through disk diffusion method, and minimum antibacterial concentrations experiments by double dilution method. The anti- microbial effects for 10 kinds of microbes by extracts of Clausena lansium leaves were studied. The results showed that the extract respectively extracted by ethyl acetate, ethanol and acetone had evidently antibacterial effect on testing bacteria (Staphylococcus aureus, Escherichia coli, Bacillus subtilis, Staphylococcus albus, Quad micrococcus and Bacillus cereus). The minimum inhibitory concentration (MIC) of ethyl acetate extract on Escherichia coli and Staphylococcus albus were on 3.2 mg/mL, on Staphylococcus aureus Bacillus cereus and Bacillus subtilis were 6.3 mg/mL, on Quad micrococcus 12.5 mg/ mL. The temperature, pH value of ethyl acetate extract had great influence on antimicrobial effects, while lighting effect was not obvious. But all solvents extraction had no obvious antibacterial effect on testing fungi (Aspergillus nigar, Penicilli- um, Rhizopus stolonifer, Candida albicans ).
