首先介绍了原子力显微镜(AFM)在ATP合成酶表面超精细结构表征中的应用,AFM同其它生化技术相结合研究ATP合成酶结构细节的相关工作,在ATP合成酶超分子表面的粘弹性、电荷分布、磁畴分布、分子间相互作用等物理属性研究中的试验进展;总结了AFM在ATP合成酶分子功能研究和单分子操纵中的应用情况;最后,综合上述试验进展现状,对仪器的机械改进和理论发展及在蛋白分子研究中的应用前景进行了讨论。
Firstly introduced the applications of AFM to the surface structure of molecular motor, photo-system complex Ⅰ and Ⅱ, membrane channels and the chaperon and so forth, and following with the application in the details of various proteins molecules. Secondly, the review stressed on the studies in physical properties of single protein molecular such as surface viscidity and flexibility, surface charge and magnetic domain distribution, intermolecular interaction,and then summarized the work on proteinst function and manipulation with some typical examples and statistic analysis. At last analyzed the dialectic relationship between physical techniques and the improvement of science in history. And ended with the conclusion that it is necessary to improve and manage the possible instruments effectively and sufficient for the personalities, and given out the optimum research routes.
