为寻求制作大熊猫肠道微生态制剂的益生菌,采用高温水浴法从20份成年健康大熊猫粪样中分离得到7株芽孢杆菌,结合细菌形态学、生理生化特性和16S rDNA基因序列分析对其进行鉴定,结果显示,分离菌分别为枯草芽孢杆菌(YS1、YY1、SY1、GG1)、蜡样芽胞杆菌(ZX1)和短小芽孢杆菌(FF1、FF2);通过对菌株抗逆性、体外抑茵性及纤维分解特性进行初步探究,证实首次培养得到了大熊猫源短小芽孢杆菌和枯草芽孢杆菌,菌株YS1、YY1、GG1、SY1耐高温能力强,ZX1、GG1耐酸能力强,绝大部分菌株对致病性大肠杆菌、沙门菌具有良好的体外抑制能力,均具有纤维素降解活性。推荐将菌株ZX1、FF2作为大熊猫益生菌制剂的候选菌株。
To research the probiotics Bacillus strains for giant pandas, 7 Bacillus strains isolated from 20 giant pandas' feces were identified by morphology, physiological and biochemical test and sequence anal- ysis of the 16 S rDNA gene. The results showed that 4 Bacillus subtilis strains(YS1 ,YYI ,GG1 ,and SY1), 2 Bacillus pumilus strains (FF1 and FF2) and 1 Bacillus cereus strain (ZX1) were obtained. The giant panda-derived B. subtilis and B. pumilus were isolated and cultivated for the first time. The stress resis- tance,cellulolytic activity, and in vitro inhibitory ability of isolated strains were studied. All strains were cellulolytic,and most could inhibit the growth of Escherichia coli and Salmonella. The activity of B. subti- lis strains at high temperature was better compared with the B. cereus strain and the two B. pumilus strains. A better acid resistant ability was observed in the GG1 and ZX1 strains. This showed that the isola- ted strains had good resistance ability, and FF2 and ZX1 could be considered as potential probiotics Bacillus for giant panda.
