为了对复齿鼯鼠粪便中分离的贾第虫滋养体进行基因型鉴定,采用饱和蔗糖溶液漂浮法进行卵囊收集,然后提取虫卵的DNA,采用巢式PCR对贾第虫的β-giardin和tpi基因位点进行扩增、测序,建立了系统进化树并进行分析。结果显示,共检测到3株贾第虫分离株,其中HNSMX03分离株为新亚型,被命名为B15,HNSMX20分离株为蓝氏贾第虫的B14亚型和E型混合感染。HNSMX49分离株为蓝氏贾第虫A型。本研究是我国首次报道复齿鼯鼠感染贾第虫,新基因亚型B15的发现为进一步研究我国野生啮齿类动物的流行病学提供了一定的资料。
To identify the genotype of Giardia strain isolated from the faeces of Trogopterus xanthipes,parasite oocysts were collected through the Sheather's sucrose flotation technique. Genomic DNiwas extracted and subjected for amplification of the β-giardin and tpi genes of giardia by nested PCR and sequencing. Phylogenetic analysis was used to identify the species and subtypes. Results showed that giardia was detected in oocysts from three T. xanthipes, i new subtype was found from HNSMX03 and named as subtype B15. HNSMX20 was mixed by subtype B14 and subtype E. HNSMX49 belonged to genotype A. This is the first report that giardia are detected in T. xanthipes,and a new subtype B15 is identified. The study provides new data for the study of the epidemiology of wild rodents in China.
