Objective：To establish a method for Determination of Allantion in Dioscorea Opposita and Rhizoma Dioscoreae and determination content of allantion.Method：separation and determination of allantion were accessed by using phenomenex Gemini 5u C 18 110A（250 × 4.6ram ） column and a mobile phase of methanol-water at a ratio of 1 ： 99（v/v）.The flow rate was 1.0mL/min,temperature at 30 ℃ and the detection wavelength at 212nm.Result：A good linearity of allantion in the rang of 2-50ug,r=0.9996;the mean value of recovery is 96.33%,RSD is 1.32%（n=6）. Conclusion：The established method is stable,simple ,accurate with good recovery and can provide an evidence for quality control of Dioscorea Opposita.
