目的:建立测定山药饮片及麸炒山药中尿囊素的方法,测定山药饮片及麸炒山药中尿囊素的含量。方法:采用phenomenex Gemini 5u C18110A(250×4.6ram)为色谱柱,流动相为甲醇-水(1:99),流速为1.0mL/min,柱温为30℃,检测波长为212nm。结果:该方法在2-50ug/mL范围内与峰面积呈良好的线性关系,r值为0.9996;加样回收率为96.33%,RSD为1.32%(n=6)。结论:该方法条件稳定,操作简单,精密度与回收率也较好,结果准确,可靠,能为山药饮片的质量评价提供依据。
Objective:To establish a method for Determination of Allantion in Dioscorea Opposita and Rhizoma Dioscoreae and determination content of allantion.Method:separation and determination of allantion were accessed by using phenomenex Gemini 5u C 18 110A(250 × 4.6ram ) column and a mobile phase of methanol-water at a ratio of 1 : 99(v/v).The flow rate was 1.0mL/min,temperature at 30 ℃ and the detection wavelength at 212nm.Result:A good linearity of allantion in the rang of 2-50ug,r=0.9996;the mean value of recovery is 96.33%,RSD is 1.32%(n=6). Conclusion:The established method is stable,simple ,accurate with good recovery and can provide an evidence for quality control of Dioscorea Opposita.
