目的:筛选土家药延龄草中抗SH-SY5Y细胞谷氨酸(Glu)损伤的有效部位。方法:Bliss法测定延龄草不同部位提取物的1%致死量(LD1)和半数致死量(LD50),建立Glu损伤SH-SY5Y细胞模型,筛选延龄草中抗SH-SY5Y细胞Glu损伤的有效部位,以形态学、细胞存活率和LDH漏出率为指标评价延龄草有效部位对Glu损伤细胞的保护作用。结果:TTM1为抗SH-SY5Y细胞Glu损伤的有效部位,能显著改善Glu损伤细胞的形态,5.0、10.0、20.0μg/mL的TTM1提高损伤细胞的存活率(P〈0.05,P〈0.001),2.5、5.0、10.0、20.0μg/mL的TTM1可显著降低损伤细胞的LDH漏出率(P〈0.05,P〈0.001)。结论:延龄草TTM1对Glu损伤的SH-SY5Y细胞具有保护作用。
Objective: To screen the effective parts of Tujia medicine birthroot against the damage of glusate(Glu) in SH-SY5Y cells. Methods: Bliss method was used to test the LD1 and LD50 of birthroot's dif erent extractives. The SH-SY5Y cells model induced by Glu was established, and effective parts against the Glu damage in SH-SY5Y cells of birthroot were screened. The protective effect of the effective part of birthroot was evaluated by the survival rate, LDH leakage rate and morphology changes. Results: TTM1 was the effective part of birthroot, which could protect the SH-SY5Y cells against Glu injury. The survival rate of damaged cells could be improved by 5.0, 10.0 and 20.0μg/mL TTM1(P〈0.05, P〈0.001). The LDH leakage rate of damaged cells could be reduced signii cantly by 2.5, 5.0, 10.0 and 20.0μg/mL TTM1(P〈0.05, P〈0.001). Conclusion: TTM1 of birthroot could protect the SH-SY5Y cells damaged by Glu.
