目的:明确天珠散介导的大鼠基因表达差异,阐释其抗血管性痴呆(VD)机制。方法:文章利用全基因组芯片检测技术确定差异表达基因,在数据库中查找差异基因表达的蛋白,通过中药语法系统推理规则,建立差异基因表达蛋白与VD相关靶点的生物网络,并推断两者间的相互作用关系。结果:通过给药组与模型组比较,得到差异表达基因229条,其中上调基因(〉2)219条,下调基因(〈0.5)10条。通过对生物网络的分析得到天珠散抗VD的3条作用途径。结论:天珠散通过介导大鼠基因表达产生差异并通过影响差异基因表达蛋白下游的生物网络发挥抗VD的作用。
Objective: To determine the gene expression difference of rats and elucidate the anti-vascular dementia mechanism mediated by Tianzhu Powder. Methods: The differential expression genes were determined by using genome-wide chip detection technology, and the expression proteins of the differential expression genes were searched in database, then the biological network between protein of the differential expression genes and vascular dementia related targets was established by the inference rule of directed TCM entity grammar system (dTGS), and the interaction between them was inferred. Results: There were 229 differentially expressed genes which including 219 up-regulated genes (〉2) and 10 down-regulated genes (〈0.5). There were 3 anti-vascular dementia pathways of Tianzhu Powder by analyzing biological network. Conclusion: By mediating the gene expression of rat and affecting the biological network of the differential expression gene downstream, Tianzhu Powder plays the role of anti-vascular dementia.
