目的通过D-半乳糖连续注射诱导树闙代谢紊乱建立衰老模型,评价脐带间充质干细胞输注对衰老树闙代谢的影响。方法 12只雌性树闙分为空白组(2只)和模型组(10只),模型组每天颈背部皮下注射10%的D-半乳糖2.5 ml/(100 g·d),空白组皮下注射等体积注射用水,连续8 w后,检测两组外周血中超氧化物歧化酶(SOD)、还原型谷胱甘肽(GSH-PX)和丙二醛(MDA)的含量变化;建立衰老树闙模型后,经尾静脉输注雄性树闙脐带间充质干细胞2次,2 w后对树闙外周血中SOD、GSH-PX和MDA的含量进行测定。结果连续注射D-半乳糖8 w后,模型组树闙体重下降,血浆SOD和GSH-PX的活性降低,MDA含量明显升高(P〈0.05)。输注树闙脐带间充质干细胞后,血清SOD和GSH-PX活性升高,MDA含量明显降低(P〈0.05)。结论连续注射D-半乳糖可诱导树闙出现衰老的特征性代谢改变,输注树闙脐带间充质干细胞对衰老树闙的代谢具有改善作用。
Objective To establish the aging model through metabolic disorder of tree shrew induced by continuous injection with D-galactose, and to evaluate the effects of umbilical cord mesenchymal stem cell infusion on the metabolism of aging tree shrew. Methods Twelve female tree shrews were divided into blank group (two) and modeling group (ten). The modeling group received subcutaneous injection in the nape with 10% D-galactose 2.5 ml/(100 g·d). The blank group received the subcutaneous injection with the same volume of water. After continuous 8 weeks, detection was made in the content changes of SOD, GSH-PX, and MDA in the peripheral blood of both groups. After the establishment of aging tree shrew model, the male tree shrews received twice the intravenous injection via the tails with umbilical cord mesenchymal stem cells. Two weeks later, detection was made in the SOD, GSH-PX, and MDA contents in the peripheral blood, Results After injection with D-galactose for continuous 8 weeks, the weight of modeling tree shrews decreased. The activity ofblood plasma SOD and GSH-PX decreased, and the content of MDA significantly increased (P 〈 0.05). After the injection with umbilical cord mesenchymal stem cells, the tree shrews' serum SOD and GSH-PX activity increased, and the content of MDA significantly decreased(P 〈 0.05). Conclusion Continuous injection with D-galactose can induce the distinctive aging metabolic changes in tree shrews. The infusion with umbilical cord mesenchymal stem cells can improve the metabolism of aging tree shrews.
