本实验从绵羊第6号染色体随机选取8个微卫星基因座,通过PCR-SSCP技术对308只南东杂交羊个体基因组DNA进行检测,分析其微卫星多态性及分子遗传学参数。结果表明:在南东杂交羊群体第6号染色体的8个微卫星标记中,共发现64个等位基因,其中基因座BMS2460、OAREL03等位基因数较少,分别为3个和2个;对8个微卫星基因座进行了分子遗传学分析,南东杂交羊的有效等位基因数(E)在1.1872~7.9431,平均有效等位基因数达到4.8119,其中MCM214的有效等位基因数最高为7.9431,BMS2460最低为1.1872;8个微卫星标记中平均杂合度(H)为0.6040,其中BM415的杂合度最高为0.8438,MCM 214最低为0.1246;多态信息含量(PIC)MCM214基因座最高为0.8633,BMS2460最低为0.1516,平均多态信息含量为0.6634说明南东杂交羊群体属于多态信息含量较高的遗传群体。南东杂交羊群体具有丰富的遗传多样性,其第6号染色体上MCM53、BMS360、BM4621、BM415、MCM2145、MCM1406个微卫星基因座可作为理想的分子标记,用于优质肉羊杂交选育过程中遗传多样性的分析。
A total of 8 random microsatellite DNA loci on sheep chromosome 6 were selected and PCR-SSCP was carried out in to analyze the polymorphisms and genetic parameters 308 South-eastern hybrid sheep population, were analyzed. The results showed that 64 alleles were found among 8 microsatellite markers on chromosome 6. Among these loci, locus BMS2460 and locus OAREL03 had fewer alleles, i.e. 3 and 2 alleles respectively. Molecular genetic analysis showed that the effective number of alleles (E) in southeast hybrid sheep was between 1.1872 and 7.9431,and the average effective number of alleles reached 4.8119. Allele MCM214 had the highest number of 7.9431, while BMS2460 shared the minimum of 1.1872. The mean heterozygosity (H) of 8 mierosatellite markers was 0.6040 with the heterozygosity of BM415 up to 0.8438, while MCM214 showed the lowest heterozygosity of 0.1246. The polymorphic information content (PIC) of MCM214 locus was 0.8633, and locus BMS2460 has the minimum of 0.1516. The average polymorphism information content was 0.6634, which indicated that southeast hybrid sheep populations belonged to high polymorphic information content genetic groups. Southeast hybrid sheep population had a rich genetic diversity, and 6 microsatellite loci on chromosome 6 (MCM53, BMS360, BM4621, BM415, MCM214 5, MCM140) and can be used as ideal molecular markers for high-quality sheep hybrid breeding process.
