内源性抗原被宿主的APC类细胞加工及提呈后,以抗原肽-MHC-I类分子复合物的形式表达于细胞表面,供特异性CD8+T细胞识别。然而,一些天然CTL表位与MHC-I的亲和力较弱,也容易引起机体的免疫耐受,并且在体内易受蛋白酶的攻击水解。针对这些不足,国内外许多学者采用了不同的方法对天然CTL表位进行改造。本文就近些年来天然CTL表位改造的主要方法做一简单的综述。
As we know that the endogenous antigens are processed and presented by the cells of host APC,then the epitope bind to major histocompatibility complex-I(MHC-I),which are then presented on the cell surface for the cytotoxic T cells(CTL) identification.However,some natural CTL epitopes,which are weakly bind to the MHC-I molecular,are likely to cause tolerance and vulnerable to hydrolysis of the protease in vivo.For the defects above,many domestic and foreign scholars have designed different methods to modify natural CTL epitope.The paper review the methods of modification on natural CTL epitope in the past several years.
