目的探讨三七总皂苷对阿霉素肾病大鼠肾组织内皮素-1蛋白及mRNA表达的影响。方法将45只大鼠随机分为5组,正常组、假手术组、模型组、苯那普利组和三七总皂苷组,手术切除大鼠右侧肾脏并腹腔注射阿霉素建立模型,10周末处死大鼠,无菌取肾脏,进行肾组织HE染色及masson染色,观察肾组织病理形态学改变,采用免疫组织化学及原位杂交方法分析肾组织内皮素-1蛋白及mRNA的表达。结果模型组大鼠肾组织肾小管上皮细胞(TEC)、肾小球系膜细胞(GMC)及肾小球上皮细胞(GEC)胞浆中ET-1蛋白及mRNA表达强阳性,三七总皂苷组大鼠肾组织ET-1蛋白及mRNA表达较模型组显著减少,有统计学意义(P〈0.05)。结论三七总皂苷能够减少阿霉素肾病大鼠肾小管上皮细胞、肾小球系膜细胞及肾小球上皮细胞胞浆中内皮素-1蛋白及mRNA的表达,进而起到保护肾功能、延缓慢性肾衰竭的病程进展。
Objective To investigate the influence of panaxnotoginsenosides on the expressions of renal endOthelin-1 ( ET-1 ) protein and mRNA in rats with adriamycin nephropathy. Methods All 45 rats were randomly divided into the normal group, sham-operation group, model group, Benazepril group and panaxnotoginsenosides group. The model was established by removing the right kidney of rat and injecting adriamycin intraperitoneally. In the end of 10^th week the rats were killed and then their renal tissue were taken out aseptically. After the renal tissues were stained by HE and Masson methods the pathomorphological changes of renal tissue were observed. The expressions of ET-1 protein and mRNA in renal tissue were analyzed by immunohistochemistry and in situ hybridization. Results In the model group the expressions of EF-1 and mRNA in renal tubular epithelial cells (TEC), glomerular mesangial cells (GMC) and glomerular epithelium cells (GEC) were strongly positive. The expressions of ET-1 and mRNA in the panaxnotoginsenosides group were decreased obviously with a statistical significant (P 〈 0.05) compared with those in the model group. Conclusion Panaxnotoginsenosides can inhibit the expressions of ET-1 and mRNA in TEC, GMC and GEC in rats with adriamycin nephropathy, protect the kidney and postpone the development of chronic renal failure.
