采用荧光光谱法研究了在pH=6.3的Na:HP04一NaH2PO。缓冲体系中壳聚糖与溶菌酶的相互作用。实验结果表明,壳聚糖对溶菌酶的荧光猝灭为静态猝灭和动态猝灭同时发生的复合猝灭,测定了不同温度下的猝灭常数。由热力学参数方程,推断出壳聚糖与溶菌酶之间的作用力主要来自疏水作用力和静电引力。运用Ftister非辐射能量转移原理,计算了壳聚糖与溶菌酶色氨酸残基间的距离为1.47nm。
The interactions between chitosan (CS) and lysozyme (LYSO) in pH = 6. 3 and Na2HPO4 - NaH2PO4 buffer is studied using fluorescence spectra method. The results indicate that the fluorescence quenching of LYSO caused by CS is a compound quenching procedure based on a dynamic and static process. The quenching constants under different temperatures are determined. There exist mainly the hydrophobic and electrostatic interactions between CS and LYSO deduced from the thermodynamic parameter equation. According to non-radiation Foster energy transfer principles, the action distance between CS and Ser residue of LYSO is 1.47 rim.
