应用荧光定量PCR及基因芯片技术,检测小分子RNA(microRNA,miRNA)在SPRN0/0型小鼠与C57BL/6野生鼠大脑中的差异表达,探讨SPRN基因敲除后小鼠大脑中miRNAs相对表达变化,对检测部分差异表达的miRNAs进行验证。结果表明:与正常标本相比,SPRN0/0型小鼠大脑中miR-135a、-24-2*、-146a、-7a、-380、-448、-128和-152的表达量均显著下调,其中miR-24-2*、-380、-448、-152均下调1/10以上,miR-135a、-146a、-7a均下调1/2以上。miRNAs的这些变化可能与Shadoo蛋白的生理功能有关。
The purpose of this study was to identify differential expressions of miRNAs between cerebrum of SPRN0/0 mice and C57BL/6 wild mice,and investigate relative expression results of miRNAs in cerebrum of SPRN0/0 mice.Some differentially expressed miRNAs analyzed with gene chips were confirmed by quantitative real-time PCR.Compared with cerebrum of C57BL/6 wild mice,eight microRNAs decreased significantly: miR-135a,-24-2*,-146a,-7a,-380,-448,-128 and-152.Four microRNAs including miR-24-2*,-380,-448 and-152 fold changed to more than 10.0,and three microRNAs including miR-135a,-146a and-7a fold changed to more than 2.0.The results demonstrate that altered miRNAs expression may be involved in Shadoo protein function and the establishment of target genes of miRNAs may provide new ideas for studying Shadoo protein function.
