选用10种含不同浓度2,4-D、Dicamba及2,4-D与Dicamba组合的诱导培养基和3种含0.01mg.L-1 2,4-D和不同浓度ZT的分化培养基,研究Dicamba、2,4-D、ZT对愈伤组织诱导和分化的影响。结果表明:2,4-D与Dicamba组合的诱导激素在再生分化上显著优于2,4-D和Dicamba。以添加1.5mg.L-1 2,4-D和0.5mg.L-1Dicamba为诱导培养基,分化培养基添加1mg.L-1 ZT和0.01mg.L-1 2,4-D对愈伤组织分化效果为最佳,绿苗分化率最高可达90%。分化培养基中1mg.L-1 ZT有利于根的分化。该研究结果为建立高效、稳定的新疆主栽小麦品种的遗传转化系统奠定了基础。
The ten induction media containing 2,4-D and Dicamba with different concentrations and a combination of both,and three differentiation media containing 0.01 mg·L-1 2,4-D and ZT with different concentrations had been selected to examine the effect of Dicamba,2,4-D and ZT on the induction and differentiation of the callus.The result showed that the induction auxin with the 2,4-D and Dicamba in regeneration and differentiation was significantly better than 2,4-D or Dicamba alone.As for the induction media,containing 1.5 mg·L-1 2,4-D and 0.5 mg·L-1 Dicamba and differentiation media containing 1 mg·L-1 ZT and 0.01 mg·L-1 2,4-D had the highest callus differentiation frequency of 90%.The addition of 1 mg·L-1 ZT in differentiation media is good to the differentiation of the root.This study is expected to lay a foundation for the establishing efficient and stable genetic transformation system for Xinjiang wheat variety.
