以五年生人参茎为供试材料,分别于人参展叶期、开花期、结果期、果后参根生长期、枯萎期采集人参茎样品,采用中性缓冲液制备粗酶液,应用分光光度法测定硝酸还原酶(NR)、超氧化物歧化酶(SOD)、谷丙转氨酶(GPT)、葡萄糖磷酸异构酶(GP!)、延胡索酸酶(FUM)的活力。结果表明:人参生长发育过程中,茎内NR和FUM均在展叶期和开花期出现活力高峰,SOD、GPT、GPI的活力在结果期较高。
Using five-year old ginseng stems as materials, changes of nitrae reductase (NR), superoxide dismutase (SOD), glutamic-pyruvic transaminase (GPT), glucose phosphate isomerase (GPI) and fumarase(FUM) activity of ginseng stems during growth and development were studied. Ginseng stem samples were collected respectively at the spreading stage, the flowering stage, fruit setting stage, tile root growing after fruit stage and the withering stage. Neutral buffer solution was adopted to extract the enzyme solution of radix ginseng. Spectrophotometry was used to test the activities of NR, SOD, GFF, GPI and FUM. The results showed that in the growth and development process of ginseng stems, the activity of NR and FUM have the peaks of the activity during the speading stage and the flowering stage. The activity of SOD, GPT and GPI during the fruit setting stage is higher than at other stages. Activity changes of 5 enzymes can provide a theoretical basis for the optimization of ginseng cultivation.
