目的 探讨芪黄明目胶囊对2型糖尿病小鼠视网膜细胞凋亡的影响.方法 KK/Upj-Ay小鼠随机分为模型组、芪黄明目高、中、低剂量组,另设C57BL/6小鼠对照组.灌胃给药3个月,取小鼠视网膜,流式细胞术测定细胞凋亡率;Real Time PCR和Western印迹法测定视网膜B细胞白血病/淋巴瘤相关抗原2 (Bcl-2)、白血病/淋巴瘤相关抗原相关X蛋白(Bax)、半胱氨酸天冬氨酸蛋白酶3(Caspase-3)、凋亡相关因子(Fas)、Fas受体(FasL)mRNA和蛋白的表达.结果 芪黄明目能不同程度的降低视网膜细胞凋亡率;降低视网膜Bax和Caspase-3的表达,升高Bcl-2表达水平及Bcl-2/Bax比值,但对Fas和FasL表达无影响.结论 芪黄明目可通过上调Bcl-2表达、下调Bax、Caspase-3表达,抑制细胞凋亡率而发挥糖尿病视网膜的保护作用.
Objective To investigate the effects of Qibuangmingmu capsule (QHMM) on retinal cell apoptosis in diabetic mice. Methods KK/Upj-Ay mice were randomly divided into model, QHMM (low, middle, high) groups. C57BIJ6 mice were selected as control group. Mice were given intragastrically for 3 months. Retina of mice was taken. Apoptotic rate was detected by FCM. Real Time PCR and Western blot were used to assess Bcl-2, Bax, Caspase-3, Fas, FasL mRNA and protein expression in retina. Results Apoptotic rate of retina, the expression of Bax and caspase were lower in QHMM group than those of model mice. The expression of Bcl-2, the ratio of Bcl -2/ Bax were higher in QHMM group than those of model mice. The expression of Fas and FasL were not obviously changed. Conclusions QHMM inhibits cell apoptosis by up-regulating Bcl-2 expression and down-regulating Bax and caspase-3 expression in retina of diabetic mice, then has protective effects on diabetic retinopathy.
