目的探讨SOX5、SOX6基因在豚鼠耳蜗中的表达及该基因对耳蜗发育的影响与作用。方法活体解剖取出豚鼠耳蜗、肝脏、脑、肾脏和心脏等组织,应用Primer 5.0软件设计SOX5、SOX6引物。使用逆转录-聚合酶链反应(reverse transcription-polymerase chain reaction,RT—PCR)法检测SOX5、SOX6基因的表达情况。结果SOX5基因在不同组织中均有表达,耳蜗中的表达相对较弱。SOX5和SOX6基因均在耳蜗组织表达。结论SOX5、SOX6基因的表达具有一定的组织特异性,两者均在耳蜗中的表达提示SOX5、SOX6基因对耳蜗的骨性组织发育可能存在一定作用。
Objective To study the effects of SOX5 and SOX6 genes on the development of cochlea and their expression in guinea pig cochlea. Methods Primers of SOX5, SOX6 were produced by Primer 5.0 software. The tissue of cochlea, brain, heart,liver and kidney of guinea pigs was obtained by vivisect. The expression of SOX5 ,SOX6 was detected by reverse transcription - polymerase chain reaction. Results SOX5 gene was expressed in all the experimental tissue with relatively lower expression in cochlea and kidney. SOX5 and SOX6 were both expressed in guinea pig cochlea. Conclusion The expression of SOX5 and SOX6 gene has definite tissue specificity. It demonstrates through the expressions in cochlea that SOX5 and SOX6 genes maybe contribute to the development of the bony structure in cochlea.
