目的:探讨不同浓度的芬太尼对食管鳞癌细胞株Eca109增殖和凋亡的影响。方法:选用不同浓度的芬太尼(0.5、5、50、500ng/ml)干预细胞。采用四甲基偶氮唑蓝(MMT)法检测细胞增殖,流式细胞仪Annexin V/PI双染色法测定细胞凋亡。结果:各组芬太尼孵育Eca109细胞24h后,对细胞的增殖无明显影响;孵育48h后,与对照组相比,各浓度组均能显著抑制细胞的增殖(P〈0.05),且呈浓度和时间依赖性。芬太尼孵育Eca109细胞48h,与对照组相比,各浓度组均能诱导早期凋亡和晚期凋亡,呈浓度依赖性(P〈0.05)。结论:芬太尼可抑制食管鳞癌细胞株Eca109的增殖并诱导凋亡。
Objective: To investigate the effects of fentanyl on proliferation and apoptosis in esophageal squamous cell carcinoma cell line Eca109. Methods: Eca109 cells cultured to logarithmic phase were randomly treated with different dose of fentanyl( 0. 5,5,50,500 ng/ml). MTT was applied to test the effect on cell proliferation and Annexin-V/PI assay was used to study its effect on apoptosis. Results: MTT test revealed that fentanly had no effect on proliferation at 24 h,but inhibited cell proliferation at 48 h and 72 h in a dose and time department manner. Annexin-V/PI assay demonstrated that fentanly induced more early and late apoptosis in Eca109 cells with increased drug dose.Conclusion: Fentanyl inhibited cell proliferation and induced apoptosis in ESCC cell line Eca109.
