目的探讨缺氧诱导因子-1α(hypoxia inducible factor 1 alpha,HIF-1α)和ERBB4在人食管鳞癌(esophageal squamous cell carcinoma,ESCC)组织中的表达,并分析两者表达的临床病理意义以及两者的相关性。方法采用免疫组化SP法检测65例ESCC组织和50例癌旁组织HIF-1α与ERBB4的表达,分析其与临床病理特征的关系以及两者表达的相关性。结果HIF-1α与ERBB4在ESCC组织中的表达的强阳性率分别为83.1%,76.9%,明显高于癌旁组织(30%和24.7%)(P〈0.05);且在分化程度较低、有淋巴转移以及TNM分期较晚的患者中,HIF-1α和ERBB4呈现显著高表达(P〈0.05);而与患者的年龄、性别、是否吸烟、是否饮酒、病变长度以及病变位置均无关(P〉0.05);HIF-1α表达与ERBB4表达之间具有相关性(r=0.629,P〈0.05)。结论 HIF-1α与ERBB4在ESCC中高表达,提示HIF-1α/ERBB4信号通路可能在ESCC发生发展中发挥重要作用。
Objective To explore the expression and clinical significance of HIF-1α and ERBB4 in esophageal squamous cell carcinoma( ESCC). Methods Immunohistochemistry( SP assay) was used to detect the expression of HIF-1α and ERBB4 in 65 patients with ESCC and 50 non-tumor adjacent tissues( NAT). The relationship between their expression and clinical features was analyzed,and their correlation was also demonstrated. Results The positive expression rate of HIF-1α and ERBB4 was 83. 1% and 76. 9% in ESCC,respectively,significantly higher than that of NAT( 30. 0% and 24. 7% respectively,P〈0. 05). And over-expression of HIF-1αand ERBB4 was correlated with tumor differentiation,lymph node metastasis and TNM stage( P〈0. 05),but showed no statistical associated with sex,age,drink,smoke,tumor length,and location. The expression of HIF-1α was positively correlated with expression of ERBB4( r = 0. 629,P〈0. 05). Conclusion HIF-1α and ERBB4 are over-expressed in ESCC,implicating that HIF-1α / ERBB4 signal pathway might play important roles in the ESCC development.
