目的:探讨YAP及磷酸化YAP在胃腺癌组织中的表达及其与胃腺癌发生发展的关系。方法:在60例胃腺癌患者中,应用免疫组化检测YAP及磷酸化的YAP蛋白在胃腺癌组织、癌旁正常组织中的表达情况。利用siRNA抑制细胞中YAP蛋白的表达,MTT检测胃腺癌细胞的增殖能力。结果:YAP蛋白可同时表达于细胞浆及细胞核中,而磷酸化的YAP主要表达于细胞浆中。与正常胃组织相比,胃腺癌组织中YAP蛋白的表达明显升高(P〈0.05),而磷酸化的YAP却显著减少(P〈0.05)。胃腺癌YAP的表达与患者性别、年龄、肿瘤的分级及分期无相关性,而与胃癌瘤体大小相关(P〈0.05)。利用siRNA抑制YAP表达可显著降低胃腺癌细胞的增殖能力。结论:胃腺癌中YAP的高表达及磷酸化YAP的低表达可能参与了胃癌的发生发展。
Objective:To explore the expressions and significance of Yes -asociated protein(YAP) and phospho- rylated YAP in gastric adenocarcinoma. Methods:Human gastric adenocarcinoma tissues and pericarcinomatousc nor- mal gastric tissues were collected and the protein expression of YAP and phosphorylated YAP(pYAP) were evaluated using immunohistochemistry. Then, human gastric cancer SGC -7901 cells were transfected with YAP- siRNA( shR- NA). The expression levels of YAP were detected by Western Blots. Effects of siRNA - mediated knockdown of YAPon cell proliferation were assessed by MTF. Results: The expression of YAP was observed in both cytoplasm and nu- cleus ,while pYAP was observed only in cytoplasm. The expression of YAP was higher in gastric adenocarcinoma than in the normal tissues (P 〈 0.05). However, pYAP in gastric adenocarcinoma was lower(P 〈 0.05 ). The YAP overex- pression was associated with tumor size, but not other clinical characteristics. Moreover, knockdown of YAP could in- hibit the proliferation of gastric cancer cells. Conclusion : YAP and phosphorylated YAP may play a key role in the in- itiation and progression of zastric adenocarcinoma.
