目的:探讨重症肌无力(MG)患者胸腺组织中胆碱乙酰转移酶(CHAT)和烟碱型乙酰胆碱受体(nAChR)的表达。方法:采用免疫组化方法检测胸腺增生(20例)、胸腺瘤(10例)、胸腺萎缩(10例)及10例正常胸腺组织中CHAT和nAChR蛋白的表达情况。结果:不同胸腺组织中CHAT和nAChR蛋白的表达差异有统计学意义(F=16.271,15.083,P〈0.001)。与正常胸腺和胸腺萎缩组织相比,胸腺瘤和胸腺增生组织中CHAT蛋白的表达增高(P〈0.05);胸腺瘤组织中nAChR蛋白表达低于胸腺增生组织(P〈0.05);胸腺萎缩组织中nAChR蛋白的表达也低于正常对照组(P〈0.05)。结论:胸腺组织中CHAT和nAChR蛋白表达水平的改变可能是MG的发病机制之一。
Aim:To explore the significance of choline acetyltransferase(CHAT) and nicotinic acetylcholine receptor(nAChR) in the thymus tissue of myasthenia gravis(MG) patients.Methods:Immunohistochemistry was used to compare the protein expression level of CHAT and nAChR in the thymus tissue from MG patients of 3 histological types(20 patients with hyperplasia,10 patients with thymoma,and 10 patients with atrophy) and normal thymus tissue from 10 normal controls.Results:The expression levels of the CHAT and the nAChR protein in thymus tissue from the 4 groups was statistical different(F=16.271,15.083,P0.001).Compared with the control group and the atrophy group,the expression of CHAT in the thymoma group and the hyperplasia group was higher(P0.05).The expression of nAChR in the thymoma group was lower than the hyperplasia group(P0.05).The expression of nAChR in the atrophy group was lower than that in the control group(P0.05).Conclusion:Changes in expression levels of nAChR and CHAT in thymus of MG patients may be one of the pathogenic mechanisms for MG.
