目的探讨核胞浆转运蛋白α-55(karyopherin alpha-5,KPNA5)在重症肌无力(myasthenia gravis,MG)患者胸腺组织中的表达情况。方法分别利用荧光定量聚合酶链反应(fluorescence quantitative polymerase chain reaction,FQ—PCR)和免疫组化染色方法,检测MG组20例患者与对照组10名受试者胸腺组织中KPNA5 mRNA和蛋白分子的表达。结果MG组和对照组胸腺组织中,KPNA 5 mRNA的表达水平分别为(0.71±0.08)和(0.57±0.02),两者之间的差异有统计学意义(P〈0.05);KPNA 5蛋白阳性细胞均数分别为(8.99±0.49)和(5.88±0.69),差异有统计学意义(P〈0.05)。结论KPNA5分子在MG胸腺组织中表达增高,可能参与了MG的发生,是MG相关基因。
Objective To study the expression of KPNA 5 in the thymus tissues of the patients with myasthenia gravis. Methods The expressions of KPNA 5 mRNA and protein in the thymus tissue of MG and the normal control were detected using fluorescent quantitative Polymerase Chain Reaction (FQ-PCR) and immunohistochemical SP method. Resuits The expression of KPNA 5 gene at mRNA level in the thymus tissues of MG ( n = 20,0. 71±0. 08 ) was significantly higher than that of the control group ( n = 10,0. 57±0. 02) ( P 〈 0. 05 ). The number of KPNA 5 positive cells in the thymus tissues of MG ( n = 18,8.99±0. 49) was remarkably higher than that of the control group ( n = 10,5.88±0. 69) ( P 〈 0. 05 ). Conclusions There is high expression of KPNA 5 in the tissue of MG patients which may be associated with the generation and development of MG.
