目的:选择合适的胸腺组织蛋白质提取和纯化方法。方法:分别采用组织裂解液法、丙酮沉淀法和蛋白纯化试剂3种方法提取7例重症肌无力患者胸腺组织蛋白质,上样量为1mg蛋白,以17cmpI3～10固相pH梯度胶条做第一向等电聚焦,SDS-PAGE垂直电泳为第二向进行双向电泳(2-DE),考马斯亮蓝染色,图像分析软件分析电泳图谱。结果:利用蛋白纯化试剂法提取的胸腺组织蛋白2-DE凝胶图像纵条纹、横条纹以及背景都明显减少,获得蛋白点数量较其他2种方法增加(P均<0.05)。结论:采用蛋白纯化试剂法提取胸腺组织蛋白,能够获得较好2-DE凝胶图像,为胸腺组织蛋白质组学的研究打下基础。
