目的探讨创伤后应激障碍(PTSD)大鼠内侧前额叶皮质(medial prefrontal cortex,mPFC)神经元Caspase-3和Caspase-9的表达变化。方法成年健康雄性Wistar大鼠50只,随机分为连续单一刺激(singleprolonged stress,SPS)模型1d、4d、7d、14d组和对照组,应用免疫组化、免疫荧光和RT-PCR方法检测PTSD大鼠mPFC神经元Caspase-3和Caspase-9的表达变化。结果 SPS刺激后Caspase-9的表达于刺激后1d开始升高,4d达到顶峰,7d和14d逐渐下降,Caspase-3于SPS刺激后1d,4d和7d表达逐渐增多,14d出现下降。结论 SPS刺激后PTSD大鼠mPFC神经元Caspase-3和Caspase-9呈规律性过表达。
Objective To investigate the expression of Caspase-3 and Caspase-9 in medial prefrontal cortex ( mPFC ) neurons of posttraumatic stress disorder (PTSD) rats. Methods A total of 50 healthy male Wistar rats were randomly divided into 1d, 4d, 7d and 14d groups of single prolonged stress(SPS) and a normal control group, immunohistochemistry, immunofluorescence and RT-PCR techniques were used to observe and detect the changes of the expressions of Caspase-3 and Caspase-9 in the mPFC neurons. Results Compared to normal control group, Caspase-9 expression gradually increased at 1d after SPS stimulation, peaked at 4 d, then gradually decreased at 7d and 14d. Caspase-3 gradually increased at 1d, 4d, and 7d after SPS stimulation, then decreased at 14d after SPS stimulation compared with normal control. Conclusion The Caspase-3 and Caspase-9 were overexpressed in medial prefrontal cortex ( mPFC ) neurons of posttraumatic stress disorder (PTSD) rats.
