目的研究创伤后应激障碍(PTSD)大鼠蓝斑神经元细胞色素C氧化酶(CcO)的表达变化。方法应用无连续单一刺激(SPS)方法建立PTSD大鼠模型,随机分为SPS刺激后1d、4d、7d、14d组和对照组,应用电镜酶化学术和RT-PCR方法观察蓝斑神经元CcO的表达变化。结果RT-PCR法显示蓝斑神经元CcO于SPS刺激后1d开始升高,4d达到高峰,7d和14d逐渐下降,但仍高于对照组。电镜酶细胞化学法显示CcO阳性反应产物主要分布在蓝斑神经元线粒体膜,SPS刺激后可见CcO释放到胞浆中。结论SPS刺激引起CcO在PTSD大鼠蓝斑核神经元呈规律性过表达。
Objective To study the expression of cytochrome c oxidase(CcO)in the locus coeruleus neurons of posttraumatic stress disorder(PTSD)-like rats.Methods Fifty normal adult male Wistar rats were randomized into:normal control group and single prolonged stress(SPS)groups(1d,4d,7d,and 14d).Transmission electronmicroscopy(TEM) and RT-PCR techniques were used to observe and detect the expression of CcO in the locus coeruleus neurons.Results Compared to control groups,level of CcO gradually increased at 1d,peaked at 4d after SPS stimulation,and then gradually decreased at 7d and 14d after SPS stimulation,but still higher than in the control group.CcO products were found to be distributed mainly in mitochondria membrane,but more CcO products were found in cytoplasm at 4d after SPS stimulation by TEM.Conclusion SPS stimulation causes the overexpression of CcO in locus coerleus neurons regularly.
