目的分析在人胚肺成纤维细胞(HELF)中热休克蛋白47(HSP47)与转化生长因子-β1(TGF-β1)之间的关系。方法将HELF分为4组:对照组、热休克组、TGF组及联合组。采用WST-8法检测HELF细胞活力和Western blot检测各组细胞HSP47、Ⅰ型胶原(ColⅠ)、α-平滑肌肌动蛋白(α-SMA)、波形蛋白(Vi mentin)表达情况。结果 TGF-β1能促进HELF的增殖(P〈0.05);在TGF-β1作用下HELF中上述蛋白表达上调,在TGF-β1联合热休克反应的作用下上调更明显,而单在热休克反应后细胞HSP47表达显著,ColⅠ却未见明显增强(P〉0.05)。结论 HSP47参与TGF-β1促HELF胶原合成,在肺纤维化中起着重要的作用。
Objective To analyze the relationship between heat shock protein 47(HSP47) and transforming growth factor beta 1(TGF-β1) in human embryonic lung fibroblasts(HELFS).Methods HELFS were divided into four groups:the control group,the heat shock group,TGF group and the combination group.HELF cell viability was measured by WST assay,the expressions of HSP47,Collagen-Ⅰ,α-smooth muscle actin(α-SMA) and Vimentin in cells were detected by Western blot.Results The cell viability was gradually increased after HELF being treated by TGF-β1(P0.05);All proteins showed obvious expression in HELF induced by TGF-β1,increased more obviously induced by TGF-β1 combined with HSR.HSP47 was obviously expressed while Collagen-I was slightly expressed after HSR(P0.05).Conclusion HSP47 may play an important role in pulmonary fibrosis as a downstream factor regulated by TGF-β1.
