以3个安徽地方猪品种皖南黑猪、圩猪和霍寿黑猪共324头猪为研究对象,采用PCR-SSCP方法检测猪TAP1基因编码区的遗传多态性。结果表明,在皖南黑猪、圩猪和霍寿黑猪TAP1基因的第10外显子41 bp碱基处发生G〉A突变,检测到AA、GA、GG 3种基因型,其中AA基因型频率在这3个群体中分别为59.53%、41.27%和54.91%,为优势基因型;皖南黑猪、圩猪和霍寿黑猪的多态信息含量(PIC)分别为0.2991、0.3611和0.3242,在该位点均处于中度多态。卡方检验表明,皖南黑猪、圩猪和霍寿黑猪群体均处于哈迪-温伯格平衡状态(P〉0.05)。
A total of 324 pigs from Wannan black pig, Wei pig, and Huoshou black pig populations were usedto detect polymorphisms of the TAP1 gene coding sequence using PCR-SSCP method. Results showed that a G 〉A mutation occurred at 41-base-position in exon 10, resulted in three genotypes (AA, GA and GG) in the three pigpopulations. The genotypic frequencies of AA in Wannan black pig, Wei pig, and Huoshou black pig populationwas 59.53%, 41.27%, and 54.91%, respectively. Genotype AA is a dominant genotype. The polymorphism infor-mation content (PIC) was 0.2991 in Wannan black, 0.3611 in Wei pig, and 0.3242 in Huoshou black pig popula-tion, which was ranged at the medium level of polymorphism. The Z2 test indicated the Hardy-Weinberg equilib-rium in the three pig populations (P〉0.05).
