目的观察白藜芦醇能否通过抑制诱导型一氧化氮合成酶(iNOS)mRNA及血红素加氧酶-1(HO—1)mRNA的表达对慢性帕金森病(PD)6.羟基多巴胺(6.0HDA)模型大鼠产生保护效应。方法经筛选无旋转行为的SD大鼠60只,随机选20只分为假手术组及正常给药组(白藜芦醇20mg·kg^-1·d^-1);应用6—0HDA纹状体内注射制作PD模型大鼠40只,随机分为模型组和白藜芦醇10、20和40mg·kg^-1·d^-1组,每日灌胃给药1次,连续10wk。电镜下观察黑质神经元超微结构的改变,real-timePCR法检测大鼠黑质部iNOSmRNA及HO-1mRNA的表达。结果电镜结果显示白藜芦醇治疗组能改善6.OHDA导致的大鼠黑质多巴胺能神经元超微结构的损伤。与模型组比较,白藜芦醇治疗组iNOSmRNA、HD-1mRNA的表达均明显降低(P〈0.01)。结论白藜芦醇对慢性帕金森病6-OHDA模型大鼠具有保护作用,其机制与抑制iNOSmRNA和HO-1mRNA的表达有关。
AIM To observe whether resveratrol can have a protective effect on 6-OHDA induced chronic Parkinson's disease (PD) rats through inhibiting the experssion of iNOS mRNA and HO-1 mRNA. METHODS Sixty SD rats without rotational behavior were screened, and 20 of them were randomly divided into sham group and normal administration group (resveratrol 20 mg·kg^-1.d^-1). The PD model rats were induced by injecting 6- OHDA into the right striatum. Fourty valid PD rats were randomly divided into model control group and resveratrol groups (10, 20 and 40 mg.kg^-1.d^-1). All groups were given orally once daily for 10 wk. The neuron uhrastructure of substantia nigra tissues was observed by electronic microscope, the levels of iNOS mRNA and HO-1 mRNA was determined by real-time PCR. RESULTS Resveratrol could alleviate 6-OHDA induced degeneration of substantia nigra dopaminergic neurons in rat. Real-time PCR showed that resveratrol could decrease the level of iNOS mRNA and HO-1 mRNA of substantia nigra in rats (vs. model group, P 〈 0.01). CONCLUSION Resveratrol have a protective effect on 6-OHDA induced chronic PD model rats, the mechanism may be related with the inhibition of the expression of iNOS mRNA and HO-1 mRNA.
