目的研究银杏叶提取物(GBE)对人牙周膜细胞(PDLCs)增殖的影响。方法采用改良组织块培养法培养人PDLCs,取第4代人PDLCs随机分为5组;①空白对照组(无GBE);②含1mg/mlGBE;③含0.1mg/mlGBE;④含0.01mg/mlGBE;⑤含0.01mg/mlGBE。以MTT法测定GBE对PDLCs增殖的影响。结果与对照组比较1mg/mlGBE组抑制细胞增殖,0.1mg/mlGBE组在12、24、48h时均能促进细胞增殖,0.01mg/mlGBE组在48、72h时能促进细胞增殖,差异具有统计学意义(〈0.05),0.001mg/mlGBE组与对照组比较差异无统计学意义(〉0.05)。结论不同浓度GBE对人PDLCs增殖影响不同。
Objective To investigate the effect of Ginkgo biloba extract on proliferation of human periodontal ligament cells(PDLCs) cultivated in vitro.Methods The human PDLCs were cultivated by modified tissue culture method.The PDLCs cultivated in vitro were divided into 5 groups and cultivated in 5 different culture media separately which all included 10 % fetal bovine serium:①control group(no GBE);②1mg/ml GBE;③0.1mg/ml GBE;④0.01mg/ml GBE;⑤0.001mg/ml GBE.The proliferation of the human PDLCs was assyed by MTT method at 12,24,48,72h.Results Compared with control group,the GBE of 1 mg/ml has an obviously inhibitory action on human PDLCs(P0.05),while the GBE of 0.1 mg/ml and the GBE of 0.01 mg/ml have promoted the proliferation of human PDLCs at 12,24,48h and at 48,72h(P0.05).The GBE of 0.001 mg/ml couldn't improve proliferation activities of human PDLCs compared with control group(P0.05).Conclusions Different concentration of Ginkgo biloba extract have different effects on human PDLCs.
