Pulsed field gel electrophoresis（PFGE） is an effective technique of gel electrophoresis which can be used to separate large DNA.This paper discusses the solutions to the problems encountered in the process of pulsed field gel electrophoresis experimental teaching.The pulsed field gel electrophoresis experiment parameters are optimized on the basis of fully understanding the principle of PFGE.Eventually,we can use the pulse field gel electrophoresis to get the better electrophoresis bands of macromolecular DNA fragments.This will lay the foundations for a further research.
