目的:观察补肾健脾活血中药"延坤煎剂"对子宫切除术后大鼠生殖内分泌激素水平的变化的影响,探讨子宫切除术后其对卵巢功能的影响。方法:将SPF级雌性成年未孕SD大鼠随机分为正常对照组、模型组、延坤煎剂高、低剂量组和乙烯雌酚(DT)组,除正常对照组外,其余各组大鼠经腹切除子宫,保留双侧卵巢。于术后第9天起分别连续给予高、低剂量延坤煎剂和DT至42天后取材,检测血清雌二醇(E2)、卵泡刺激素(FSH)、黄体生成素(LH)、睾酮(T)水平。结果:子宫切除术后大鼠血清E2和T水平明显降低,FSH水平明显升高,差异有统计学意义(P〈0.01,P〈0.01,P〈0.05);延坤煎剂高剂量组与模型组相比,FSH水平下降,E2和T水平升高,三者两两比较,差异有统计学意义(P〈0.01,P〈0.01,P〈0.05);DT组与模型组相比,FSH水平下降,E2水平升高,两组差异有统计学意义(P〈0.01,P〈0.05)。结论:子宫切除术可使大鼠卵巢功能衰退,延坤煎剂可改善子宫切除术后大鼠的生殖内分泌功能,防治卵巢功能早衰。
Objective:To observe effects of Yankun Recipe(YKR) on the reproductive endocrine function in rats after hyster- ectomy, and to explore the influence of ovarian function after hysterectomy. Methods : Rats were randomly divided into the normal control group, the model group, the high dose YKR group, the low dose YKR group, and the DT group. The rat model Was prepared using abdominal hysterectomy. The high and low dose YKR and DT was respectively administered to rats from the ninth day to the 42 days. The sample was taken. The rats serum estradiol ( E2 ), follicle - stimulating hormone ( FSH), luteinizing hormone ( LH ) and testosterone (T) were determined by using radioimmunoassay. Results :Compared with the normal control group,the estradiol ( E2 ) and testosterone (T) were significantly decreased in the model group ( P 〈 0. 01 ). The follicle - stimulating hormone (FSH) was significantly increased(P 〈 0. 05 ). Compared with the model group, the follicle - stimulating hormone (FSH) was significantly decreased in the high dose YKR group (P 〈 0. 01 ), the estradiol( E2 )and testosterone (T) were significantly in- creased( P 〈0. 01 ,P 〈0. 05 );the follicle- stimulating hormone (FSH) was significantly decreased in the low dose YKR group ( P 〈 0. 05 ) ; the follicle - stimulating hormone (FSH) was significantly decreased in the DT group ( P 〈 0. 01 ), the estradiol ( E2 ) was significantly increased (P 〈 0. 05 ). Conclusion:Rat ovarian function declines after hysterectomy. Yankun Recipe has a regula- tion of preventing premature ovarian failure and improving reproductive endocrine function of the rats after hysterectomy.
