目的:分析ID4基因在正常组织细胞和癌变组织细胞中的表达情况,为研究该基因的表达调控机制奠定基础.方法:采用生物信息学方法,利用开放性的数据库包括Gene expression omnibus(GEO)、Affymetrix基因芯片表达数据库、人类基因组数据库(GeneBank)、基因表达数据库SAGE、GeneCard、InterPro、ProtoNet、UniProt和BLOCKS.结果:ID4基因在多种神经系统肿瘤细胞中的表达高于正常脑组织(209/3,148/3,123/3,72/3,64/3,45/3);而甲状腺滤泡癌的表达要低于正常甲状腺组织(35/172);正常乳腺组织要明显低于转移乳腺癌和原发乳腺癌的表达(10/36,10/34);正常肺组织的表达高于肺腺癌(11/4).此外,ID4基因在正常淋巴结、胚胎干细胞等细胞中也有表达.结论:生物信息学可以用来方便快捷地分析基因的表达谱;ID4基因的表达具有组织特异性.
Objective:To explore the expression series of ID4 gene in normal and malignant tissues or cells as a primary work for studying the regulation mechanism of ID4 gene. Methods: Bioinformatics and free open data-set such as GeneBank, SAGE, GeneCard, GEO, InterPro, ProtoNet, UniProt and BLOCKS were used to analyze the expression series of ID4 mRNA and ID4 protein. Results: ID4 gene had higher expression in a lot of malignant neuro-tissues and cells than in normal brain tissues (209/3, 148/3, 123/3, 72/3, 54/3, 45/3 ) ; higher in normal thyroid cells than in follicular thyroid carcinoma cells ( 172/35 ), in metastasis and in situ breast carcinoma higher than normal breast (34/10, 36/10) , in normal lung tissues than lung adenocarcinoma(11/4). Further more, ID4 gene had expression in normal bone marrow, lymphocyte and embryonic stem cell normal cell. Conclusion:Bioinformatics can be used freely to analyze a gene's expression series. ID4 gene has different expression in different tissues.
