目的探讨骨髓增生异常综合征(MDS)患者中Id4基因甲基化状态。方法采用甲基化特异性聚合酶链反应(MS-PCR)对56例不同类型MDS患者骨髓进行Id4基因启动子区甲基化状况检测,并以非恶性血液肿瘤骨髓作为对照。结果Id4基因在非恶性血液肿瘤骨髓中呈完全性非甲基化状态,在MDS各种类型中,骨髓中原始细胞比例分别低于和高于5%的两组患者中,Id4甲基化差异有显著性意义。结论骨髓原始细胞比例高的MDS患者骨髓Id4基因甲基化发生率高。
Objective To investigate the difference of Id4 gene promoter methylation in patients with different subtypes of myelodysplastic syndrome(MDS).Methods MS-PCR was used to detect the Id4 gene methylation in 56 patients with different subtypes of MDS.Id4 gene methylation was detected in bone marrow samples from healthy donors.Results No methylation of Id4 gene was found in bone marrow samples from healthy individuals.In subtype groups of MDS patients with a lower or higher ratio of blast cells than 5%,the incidence of Id4 gene methylation was different (P〈0.05). Conclusion Id4 gene methylation can be found in MDS patients with a higher ratio of blast cells.
