目的探讨烟草表达的粉尘螨I类变应原重组蛋白对小鼠的致敏性。方法40只BALB/c小鼠随机分为4组,分别为PBS阴性对照组、卵白蛋白阳性对照组、原核表达Derfl(pDerfl)组、烟草表达Derfl(tDerfl)组,各组小鼠分别于第0、7、14天腹腔注射,PBS组给予PBS,其余3组分别用卵白蛋白、pDerfl、tDerfl致敏,第21天起连续7d雾化吸入激发,24h后处死。通过肺组织病理切片观察小鼠肺部炎症,收集支气管肺泡灌洗液(BALF)进行总细胞及分类计数,以ELISA法检测BALF、脾细胞培养上清(SCCS)的特异性细胞因子IL-2、IL-4、IL-5、IL-17和IFN-1和血清中变应原特异性IgE、IgGl及IgG2a抗体浓度。结果PBS组小鼠肺部未见明显病理改变,卵白蛋白、pDerfl、tDerfl组小鼠肺部出现明显的炎症。tDerfl组小鼠BALF中的细胞总数、嗜酸性粒细胞、中性粒细胞、淋巴细胞、抗原特异性细胞因子IL-4、IL-5、IL-17和SCCS中IL-4、IL-5,IL-17均明显高于阴性对照组(P〈0.01),而与阳性对照组相比无统计学意义;tDerfl组小鼠BALF和SCCS中IL-2、IFN-1均低于阴性对照组,差异有统计学意义(P〈0.01);卵白蛋白、pDerfl、tDerfl组特异性IgE和IgGl抗体水平均高于PBS组(P〈0.01)。结论从烟草中表达的重组Derfl蛋白对小鼠具有致敏作用,其可作为变应原进一步研究。
Objective To analyze the allergenicity of the major allergen group I from Dermatophagoides farinae (Der fl) expressed in tobacco. Methods Forty BALB/c mice were randomly divided into four groups, namely PBS (negative Control), ovalbumin (positive group), prokaryotic expression Der fl (pDer fl), tobacco expression Der fl (tDer fl). PBS group were treated with PBS all the time, the other three groups were sensitized intraperitoneally with OVA, pDer fl, tDer fl respectively on day 0, 7, 14, and then inhalation method were used to challenge on day 21 for 7 days. Twenty-four hours after the last challenge, mice were sacrificed and the lungs were made into slices stained by hematoxylin and eosin in order to observe the pathologic changes in lungs. The bronchoalveolar lavage fluid (BALF) was collected to determine the number of the total cells, neutrophils, lymphoeytes and eosinophils by Liu's stain. ELISA was used to assay the level of cytokines (IL-2, IL-4, IL-5, IL-17, IFN-γ) in BALF and in the supernatant of splenocyte culture (SCCS) and the level of Der fl-speeific IgE, IgG1 and IgG2a in the sera. Results The pulmonary tissue section revealed no pathological changes in samples from PBS group, but significant allergic inflammation was found in the animals from group OVA, pDer fl, tDer fl, and the total cell number [(20.11±5.29) × 105], eosinophils [(1.83±0.65)×105], neutrophils [(4.86±1.11)×105] and lymphocytes [(8.58±1.61)×105] in BALF from tDer fl were also increased significantly as compared with PBS group. The cytokines IL-4 [(58.31±11.74) pg/ml], IL-5 [(78.11 ±7.68) pg/ml], IL-17 [(635.54±34.79) pg/ml]in BALF from tDer fl were significantly higher than PBS group (P〈0.01). The cytokines IL-4 [(78.74±11.23) pg/ml], IL-5 [(35.41±8.29) pg/ml], IL-17 [(608.64±67.90) pg/ml] in SCCS from tDer fl were also significantly higher than PBS group.
