以三油酸甘油酯为底物,研究苦荞麦多糖在不同条件下对胰脂肪酶抑制作用的影响,确定其最佳抑制条件、抑制作用类型及抑制常数。结果表明,苦荞麦多糖对胰脂肪酶具有明显抑制作用,其半抑制浓度IC50为28.23mg/m L。多糖浓度为30mg/m L,反应时间为15min,p H为7.5,温度为37℃时抑制效果最佳,抑制率达到52.17%,为非竞争性抑制,抑制常数Ki=43.28mg/m L。通过紫外吸收光谱测定表明胰脂肪酶紫外吸收光谱最大吸收波长发生红移,显示苦荞麦多糖是通过改变胰脂肪酶的构象,致使其酶催化活性受到抑制。苦荞麦多糖可以作为植物源的胰脂肪酶抑制剂发挥减肥因子的作用。
Glycerol trioleate as substrate,the bitter buckwheat polysaccharide under different conditions influence on inhibition of pancreatic lipase,determine the best conditions,the types of inhibition and inhibition of constant.The results showed that the bitter buckwheat polysaccharide has obvious inhibitory effect on pancreatic lipase,the IC50 half inhibitory concentration of 28.23mg/m L. By ultraviolet absorption spectrometry showed that pancreatic lipase maximum absorption wavelength ultraviolet absorption spectrum redshift,bitter buckwheat showed the polysaccharide is by changing the conformation of pancreatic lipase,leading to the enzyme catalytic activity has been restricted.Polysaccharide can be used as a bitter buckwheat plant source pancreatic lipase inhibitor play the role of weight loss factor.
