目的:观察survivin反义寡核苷酸(Antisense Oligodeoxynuleotide,ASDON)对人胆管癌QBC939细胞株凋亡增殖及survivin和cyclin D1表达的影响。方法:合成特异性靶向survivin ASODN,并将其转染QBC939细胞,流式细胞术观察survivin ASODN对QBC939细胞凋亡和增殖的影响;同时,应用免疫组化检测survivin ASODN对QBC939细胞中survivin和细胞周期素D1(cyclin D1)蛋白表达的影响。结果:ASODN转染组细胞凋亡指数明显高于对照组(P〈0.05),ASODN转染组细胞增殖指数明显低于各对照组(P〈0.05),ASODN组survivin和cyclin D1的蛋白表达明显低于对照组(P〈0.05)。结论:Survivin ASODN转染能诱导胆管癌细胞QBC939凋亡,抑制细胞增殖,并可下调survivin和cyclin D1蛋白表达。
Objective:To investigate the effects of survivin antisense oligodeoxynucleotides (ASODN) on apoptosis and proliferation and the effects of survivin ASODN on the expression level of survivin and cyclin D1 protein in human cholangiocarcinoma cell line QBC939.Methods:Specific target selective survivin ASODN, designed and artificially synthesized, was transfected to the cell line, flow cytometry was used to study cell proliferation and apoptosis in cell line QBC939.The expression of survivin and cyclin D1 protein were detected by immunohistochemistry.Results:Apoptosis index in ASODN group was significantly higher than that in control group (P0.05),proliferation index in ASODN group was significantly lower than that in control group (P0.05).The expression level of survivin and cyclin D1 protein in ASODN group were significantly lower than that in control group (P0.05).Conclusion:Survivin ASODN can induce apoptosis and inhibit proliferation in human cholangiocarcinoma cell line QBC939,and can inhibit the expression level of survivin and cyclin D1 protein.
