补阳还五汤对脑梗死大鼠内源性神经干细胞增值分化作用的研究

目的:研究补阳还五汤对内源性神经干细胞(NSC)归巢缺血脑组织的作用。方法:将240只雄性大鼠随机分为模型组、假手术组、补阳还五汤治疗组、正常对照组,采用线栓法制作脑梗死模型,分别给予不同处理,于术后1、3、7、14、21、28天6个时间点分批处死大鼠。用免疫组织化学方法检测各组大鼠海马区BrdU标记的阳性细胞情况和梗死区域及相对应区域的NeuN标记的阳性细胞情况。结果:正常组和假手术组大鼠的海马区可见到少量的BrdU标记阳性细胞,与模型组脑缺血区域相对应区域可见大量的NeuN标记阳性细胞,差异均无明显统计学意义(P>0.05),与假手术组相比,模型组和补阳还五汤治疗组大鼠BrdU标记阳性细胞数目增多,NeuN标记阳性细胞数目减少,差异均具有统计学意义(P<0.05),与模型组相比,补阳还五汤治疗组大鼠BrdU、NeuN标记阳性细胞数目均增多,且第7天时BrdU标记阳性细胞数目达到高峰,第14天时NeuN标记阳性细胞数目达到高峰,差异均具有统计学意义(P<0.05)。结论:补阳还五汤可以诱导脑梗死大鼠内源性神经干细胞的增殖分化,在一定程度上具有促进神经再生的作用。

英文摘要:

Objective:To observe the effect of the Buyang Huanwu decoction on NSC homing ischemia brain tissue.Methods:240clean and healthy male SD rats were divided into the model group,fake surgery group,Buyang Huanwu decoction group and normal control group.Cerebral infarction models were made by line bolt legal system;by different methods,in postoperative1,3,7,14,21,28days,rats were killed.The positive cells marked by BrdU in rat hippocampus and positive cells marked by NeuN in infarction area were checked.Results:In the normal group and fake surgery group,a small amount positive cells marked by BrdU in rat hippocampus were found.And there was a large number of positive cells marked by NeuN in the same part of ischemia brain region.But differences were not marked statistically significant(P>0.05).Compared with fake surgery group,positive cells marked by BrdU in the model group and Buyang Huanwu decoction group were increased.The number of positive cells marked by NeuN were decreased.And the differences were statistically significant(P<0.05).Compared with the model group,positive cells marked by BrdU,NeuN in the Buyang Huanwu decoction groupwere increased,and positive cells marked by BrdU increased to the top on7-day and ones marked by NeuN on14-day.And the differences were statistically significant(P<0.05).Conclusion:The Buyang Huanwu decoction can induce the proliferation of endogenous neural stemcells after cerebral infarction,and promote nerve regeneration.
