目的:研究补阳还五汤对大鼠内源性神经干细胞GFAP、SYN表达的影响。方法:将240只雄性大鼠随机分为模型组、假手术组、补阳还五汤治疗组、正常对照组,采用线栓法制作脑梗死模型,分别给予不同处理,于术后1、3、7、14、21、28天6个时间点分批处死大鼠。采用免疫组织化学方法检测各组大鼠梗死区域GFAP、SYN标记的阳性细胞情况。结果:模型组大鼠脑缺血区域相对应区域可见大量的GFAP、SYN标记阳性细胞,差异均无明显统计学意义(P〉0.05)。与假手术组相比,模型组和补阳还五汤治疗组大鼠GFAP标记阳性细胞数目增多,SYN标记阳性细胞数目减少,差异均具有统计学意义(P〈0.05)。与模型组相比,补阳还五汤治疗组大鼠GFAP、SYN标记阳性细胞数目均增多,第14天时GFAP、SYN标记阳性细胞数目达到高峰,差异均具有统计学意义(P〈0.05)。结论:补阳还五汤是治疗脑梗死恢复期的中药经验方,在改善神经功能缺损方面有显著疗效。补阳还五汤能促进脑梗死后内源性神经干细胞增殖分化为神经元、星形胶质细胞等神经组织的后备细胞,并能增强神经突触之间的联系,使神经元发挥正常的兴奋性,促进神经功能恢复。
Objective: To observe the effect that mobilization of endogenous neural stem cells homing ischemia brain tissue of Bu Yang Huan Wu soup. Methods: There are 240 clean and healthy male SD rats being divided into 4 groups including model group, fake surgery control group,Bu Yang Huan Wu soup cure group and normal control group. Using line bolt legal system for cerebral ingarction model, respectively for different processing,in postoperative 1,3,7,14,21,28 days six time point to kill rats. And to check the positive cells marked by GFAP and SYN in infarction area or homologous area. Results : There was a large amount of positive cells marked by GFAP and SYN in the same part of ischemia brain region. But differences were not marked statistical significance(P 〉0.05). Compared with fake surgery control group,positive cells marked by GFAP in model group and Bu Yang Huan Wu soup cure group rats increased. The number of positive cells marked by MBP decreased. And the differences were statistically significant (P〈 0.05). Compared with model group,positive cells marked by GFAP and SYN in Bu Yang Huan Wu soup cure group rats increased, and positive cells marked by GFAP and SYN increased to the top in the fourteenth day. And the differences were statistically significant (P〈0, 05). Conclusion: Bu Yang Huan Wu decoction is a Chinese medicine experience in treatment of cerebral infarction con- valescence, has significant curative effect in improving neurological function. The results of the study show that Buyanghuanwu decoction can promote cerebral infarction after five endogenous neural stem cell proliferation and differentiation into neurons, astrocytes and neural tissue reserve cells,and enhance synaptic connections between the neurons play normal excitability,promote the recovery of nerve function.
