目的:应用国际临床化学与检验医学联合会(IFCC)公布的参考方法对血清乳酸脱氢酶、γ-谷氨酰基转移酶及淀粉酶进行测定,其准确性通过参考实验室国际比对计划的评估。方法:按照IFCC有关酶学活性在37℃测定参考方法的标准操作程序,使用Hitachi测定系统进行3个酶的测定。通过测定质控血清监测系统稳定状态、有证参考物和参加酶学国际比对,验证参考方法的准确性。结果:本测定系统测定室内质量控制血清3种酶日间测定变异系数为0.5%~1.4%;有证参考物测定结果在允许的范围内,初步验证了参考方法的准确性;3种国际比对样本A、B两个浓度的测定值均位于所有参考实验室测定值的軃x±2s范围内。结论:应用IFCC参考方法采用本测定系统对3个酶进行的测定结果稳定、准确,具有一定的可比性。
Objective:To detect serum Lactate Dehydrogenase(LDH),γ-Glutamyltransferase(GGT) and α-amylase(AMY) by using the reference methods published by IFCC,and to evaluate the accuracy by participating in the ring trial among National Reference Laboratories.Methods:According to the IFCC primary reference procedures for the measurement of catalytic activity concentrations of enzymes at 37℃,the three enzymes were detected on the Hitachi measuring system.Accuracy of the reference methods was tested and verified by monitoring the stability of measuring system through the detection of control sera,certified reference materials(CRM) as well as participating in the ring trial.Results:With this measuring system,the intra-day variability(CV) of the three enzymes was 0.5%~ 1.4%,and the result of CRM was in the permitted measuring range,all of which exhibited the accuracy of the reference methods.Com-pared with the values from all participating reference labs,the detecting results of the three kinds of specimens for the international ring trial,each including concentration A and B,wre in the range of ±2s.Conclusions:By using the IFCC reference methods,the de-tecting results of the enzymes are stable,accurate and comparatable.
