油酸诱导急性呼吸窘迫综合征大鼠心脏局部RAS 的影响及心功能变化相关研究

目的:观察油酸诱导急性呼吸窘迫综合征(ARDS)大鼠心脏局部 RAS的影响及心功能的变化。方法雄性SD大鼠36只,随机分成假手术组和造模组,每组18只,各组又按造模成功后1h、2h和4h分为a、b、c3个亚组。(1)假手术组(S组):仅进行气管切开插管和右颈总动脉穿刺置管,尾静脉注射生理盐水0.15 mL/kg ;(2)造模组(C组):尾静脉注射油酸0.15 mL/kg ,建立油酸诱导的大鼠A RDS模型。每组有效检测与活杀取材的大鼠均为18只。结果与S组比较,C组L V-EDP、AngⅡ含量及Renin活性均明显升高(P<0.05),LVSP、±LVdp/dtmax明显下降。结论 ARDS可致大鼠心肌损害、心功能降低,同时激活心脏局部RAS。

英文摘要:

Objective To observe the relationship of the cardiac function and the level of local RAS in acute respiratory distress syndrome (ARDS) rats induced by oleic acid .Methods Sprague-Daw-ley rats were randomly divided into two groups .Group S :Control Group (only tracheal intubation ,the right carotid artery catheterization ,and tail vein injection of normal saline 0 .15 mL/kg .n=18);Group C ( ARDS group:the ARDS model of rats induced by tail vein injection of oleic acid 0 .15 mL/kg ,n=18) .Two groups were randomly divided into 3 subgroups respectively by 1 h ,2 h and 4 h after the modeling ,and there were 6 rats in each group with effective detection and killed alive for materials .Recording the left ventricular systolic pressure (LVSP) ,maximum rising and falling rate of left intraventricular pressure ( ± LVdp/dtmax) ,and left ventricular end-diastolic pressure (LVEDP) at each time point .AngiotensinⅡ( AngⅡ) concentration in myocardial tissue and renin activity level ( Renin ) were be detected respectively . Results In ARDS rats ,LVSP ,LVEDP ,± LVdp/dtmax ,Ang Ⅱcontent and Renin activity were high-er than control group with significant difference (P〈0 .05) .ConclusionARDS could induce myocardial damage ,decrease cardiac function and activation of cardiac local RAS in rats ..
